Th17 master transcription factors ROR alpha and ROR gamma regulate the expression of IL-17C, IL-17D and IL-17F in Cynoglossus semilaevis

摘要The RAR-related orphan receptors (RORs) are members of the nuclear receptor family of intracellular transcription factors. In this study, we examined the regulatory properties of ROR alpha (CsROR alpha) and ROR gamma (CsROR gamma) in tongue sole (Cynoglossus semilaevis). CsROR alpha and CsROR gamma expression was detected in major lymphoid organs and altered to significant extents after bacterial and viral infection. CsROR alpha enhanced the activities of C5IL-17C, C5IL-17D, and C5IL-17F promoters, which contain CsROR alpha and CsROR gamma binding sites. CsROR gamma also upregulated the promoter activities of C5IL-17D and C5IL-17F but not CsIL-17C CsROR alpha and CsROR gamma proteins were detected in the nucleus, and overexpression of CsROR alpha in tongue sole significantly increased the expression of C5IL-17C, CsIL-17D, and C5IL-17F, whereas overexpression of CsROR gamma significantly increased the expression of CsIL-17C and CsIL-17F but no C5IL-17D. These results indicate that ROR alpha and ROR gamma in teleost regulate the expression of IL-17 members in different manners. (C) 2015 Elsevier Ltd. All rights reserved.
